Senescence represents a fundamental biological process whereby cells irreversibly cease to divide and enter a state of permanent growth arrest, specifically at the G1 phase. Despite halting division, senescent cells remain metabolically active, releasing substances that can provoke inflammation and potentially damage adjacent healthy cells. This activity is implicated in the development of various diseases, including cancer. As cells age after a period of rapid proliferation, they slow their division rate and become resistant to stimuli that typically promote division. Senescent cells exhibit distinctive characteristics: an increase in size, flattened shape, altered gene expression patterns, and the activity of senescence-associated β-galactosidase (SA-β-gal).
The Senescence Detection Kit is an essential tool for measuring the activity of SA-β-gal in cell cultures. This kit functions by hydrolyzing X-gal (5-Bromo-4-chloro-3-indolyl β-D-galactosidase), leading to the accumulation of a blue color specifically in senescent cells.
