Secreted alkaline phosphatase (SEAP) is favored as a reporter of gene expression over traditional intracellular reporters like CAT and firefly luciferase, due to its secretion into the culture medium. This allows for non-invasive tracking of gene expression through SEAP mRNA and protein levels. Continuous sampling of the medium over time enables the study of gene expression kinetics without harming the cells, which can be used later for RNA or protein analysis. Initially, SEAP activity was detected using p-nitrophenyl phosphate; however, modern methods utilize chemiluminescent substrates like CSPD for more sensitive and rapid detection.

The ZellX® SEAP (Secreted Alkaline Phosphatase) Reporter Gene Assay Kit is designed for monitoring the activity of reporter genes that encode the SEAP enzyme. In genetic research, these kits are used to study gene expression under various experimental conditions.
