 ZELLX® High Fidelity DNA Polymerase for long PCR is an accurate and robust enzyme blend that combines Taq DNA Polymerase and a DNA proofreading Polymerase with 3’→5’ exonuclease activity which is optimized for highly accurate and efficient PCR amplification of very long DNA templates (long range PCR). It exhibits magnificent results with very long (up to 15 kb), GC-rich or other difficult templates.

Due to its inability to repair nucleotide mismatches following misincorporation, Taq DNA Polymerase is inefficient at amplification of fragments larger than 3–5 kb. The enzyme mixture includes a highly possessive 5’→3′ DNA polymerase that performs a 5’→3′ polymerization-dependent exonuclease replacement activity, and a proofreading enzyme with 3’→5′ exonuclease proofreading activity which allows mismatches to be repaired and extension to continue and results in a greatly increased fidelity of DNA synthesis (6.5-fold) compared to Taq polymerase. ZELLX® High Fidelity DNA Polymerase produces PCR products with blunt ends and generates 3´-adenine overhang in amplified DNA fragment which can be further used in cloning into T-vectors.
