TE buffer, also known as Tris-EDTA buffer, is a widely used buffer solution in molecular biology and biochemistry laboratories. It is composed of two main components: Tris (tris (hydroxymethyl) aminomethane) and EDTA (ethylenediaminetetraacetic acid). The primary purpose of TE buffer is to maintain a stable pH and prevent the degradation of nucleic acids, such as DNA and RNA.

The two main components of TE buffer serve different functions:

Tris which Acts as a buffering agent to maintain a constant pH level. It helps to resist changes in pH, even when small amounts of acidic or basic substances are introduced. And EDTA that Serves as a chelating agent that binds to and sequesters metal ions, such as magnesium and calcium. Metal ions can promote the activity of nucleases, which are enzymes that degrade nucleic acids. By sequestering these metal ions, EDTA helps to protect DNA and RNA from enzymatic degradation.

TE buffer is commonly used in various molecular biology applications, including:

DNA and RNA extraction: It is used to resuspend or dissolve DNA and RNA pellets obtained during purification procedures. DNA and RNA storage: TE buffer is employed to maintain the stability of nucleic acids during long-term storage. PCR reactions: TE buffer can be used as a diluent for DNA templates in PCR reactions. Electrophoresis: It is used as a sample buffer in agarose gel electrophoresis for the separation and analysis of nucleic acids. ZELLX® TE Buffer, for instance, is composed of 0.1 M Tris-HCl, 0.010 M EDTA with a pH of 7.4 at 25°C.
